Title: The regulatory role of macrophage in the cartilage regeneration after acute injury
Abstract: Macrophages have played an important role in regulating regeneration in various tissues, such as heart, muscle, lung, nerve, bone etc. Despite of their involvement in the pathology of arthritis, how they regulate cartilage regeneration after acute injury remains elusive, the microenvironment of which is different from that of arthritis. To understand immune regulation of cartilage regeneration, we propose the exploration of macrophage function in cartilage regeneration by: (1) developing murine osteochondral defect model with selective joint macrophage depletion to investigate the influence of macrophage depletion on autologous regeneration, (2) developing murine osteochondral defect model with macrophage-based therapy to investigate the influence of exogenous macrophage on cartilage regeneration, (3) investigating cartilage stem cells to explore possible interactions with macrophages. Macrophages can regulate cartilage regeneration in a phenotype- and time- dependent manner.
