Title: Construction of Cas9-Avd system to Enhance Homologous Recombination Efficiency

Abstract: Homologous recombination is responsible for the maintenance of genome integrity. By introducing an artificial DNA template as donor, certain exogenous sequences could be inserted into genome precisely. But the efficiency is low either in mammlian cell lines or embryos. In my proposal, a novel strategy to enhance homologous recombination efficiency is described and constructed, based on Avidin and Biotin interaction mechanism. This system works well based on primary data and shows huge potential to application.
